Nested PCR #
Nested PCR 란? #
- 다른말로 second PCR 이라고 한다.
- 1차 PCR을 한 후, 산물을 이용하여 2차 PCR을 하는 방법을 말한다.
Nested PCR 원리 #
1) 1차 PCR을 하면 primer가 증폭하려는 template에 완벽하게 binding하지 않을 경우, contamination 산물이 생길 수 있다.
2) 1차 산물로 얻어진 fragment들의 target DNA 부위를 2차 PCR을 통해 증폭하면 더 정확한 산물을 얻을 수 있다.
3) 1차 PCR을 할때, 2개의 primer를 사용 (forward primer, reverse primer)
4) 2차 PCR을 할때, 1차 primer보다 안쪽 서열로 primer를 짜고 PCR을 수행한다. (그림 참조)
5) 즉, 총 4개의 primer가 사용된다.
Nested RT-PCR #
원리는 Nested PCR과 같으며 다만 RNA를 초기 기질로 사용한다는 점에서 차이가 있음
Nested PCR 장점 #
1) PCR에서 증폭된 fragment가 non-specipic하게 증폭된 것인지 확인할 때 사용
2) 매우 적은 발현량을 갖는 RNA를 증폭할 때 쓸수 있음 (Nested RT-PCR) 3) target을 정확하게 증폭할 수 있음Nested PCR 활용 #
